The embryo and its extraembryonic membranes are transferred to a Petri dish on day 3 or 4 of incubation and CAM develops at the top as a flat membrane and reaches the edge of the dish to provide a two-dimensional monolayer onto which multiple grafts can be placed (Auerbach et al., 1974; Jakobson et al., 1989). Because the entire membrane can be seen, rather than just a small portion through the shell window, multiple grafts can be placed on each CAM and photographs can be taken to document vascular changes over time.
