Polysaccharides possess unique chemical, physical, and biological properties and are the most abundant natural biopolymer. Monosaccharide building blocks and glycosidic linkages establish the backbone of these polymers and determine the diversity and complexity of the polysaccharides. This chapter introduces the chemistry of carbohydrates, as well as the means for synthesizing defined oligosaccharides, and the properties and applications of several important polysaccharides. To afford access to well-defined polysaccharides, enzymatic, chemical, and chemoenzymatic approaches will be described. These advances continue to facilitate a growing number of exciting biomedical applications of polysaccharides by academia and industry.
